Since nitrosamine impurities were first identified in medicines, the pharmaceutical industry has relied on highly conservative approaches to protect patient safety. While appropriate, this has also resulted in extensive testing and significant resource demands.

During our recent panel discussion (▶️Watch on demand: https://ow.ly/PKoy50ZPKSN ),
toxicologist George Johnson suggested that 50–90% of today's nitrosamine testing could eventually become unnecessary if emerging scientific approaches are adopted by regulators.

📝 A recent study led by scientists at Eli Lilly and Company helps explain why. It proposes a complementary pathway in which quantitative in vivo mutagenicity data (TGR) could support the derivation of compound-specific acceptable intake limits, moving risk assessment towards a more evidence-based approach. Why do some populations experience a higher risk of to***co-related cancer despite similar smoking behaviours?

Researchers have explored this question for decades.

📝A new study led by US scientists investigates whether inherited differences in to***co carcinogen metabolism could be part of the explanation. DOI: 10.1007/s00204-026-04505-w

Focusing on NNK, a major to***co-specific nitrosamine carcinogen, the authors found that smokers with higher African genetic ancestry were more likely to exhibit a poor metaboliser phenotype. They also identified ancestry-associated genetic variants involved in to***co carcinogen metabolism and detoxification.

The findings suggest that people smoking the same number of ci******es may not receive the same biological dose of carcinogen exposure, potentially contributing to differences in to***co-related cancer risk.

✨It's a fascinating example of how biomarkers, molecular toxicology and genetics are helping us better understand individual susceptibility to carcinogens—and could ultimately support more personalised approaches to cancer prevention.
