Activated PI3K delta syndrome (APDS) is an ultra-rare genetic immune disorder caused by overactivity of a pathway that helps control the immune system. People with APDS may have repeated infections, enlarged lymph nodes or spleen, low antibody levels, poor responses to vaccines, abnormal immune cell counts, and an increased risk of certain blood cancers and they are may be first diagnosed with common variable immunodeficiency (CVID) or hypogammaglobulinemia. APDS is diagnosed through genetic testing and can be treated with immunoglobulin replacement, antibiotics, antivirals, supportive care, and, in some countries, a targeted medication called leniolisib that helps correct the overactive immune pathway.
