What are superbugs and how can I protect myself from infection?
Answers from Pritish K. Tosh, M.D.
"Superbugs" is a term used to describe strains of bacteria that are resistant to the majority of antibiotics commonly used today. Resistant bacteria that cause pneumonia, urinary tract infections and skin infections are just a few of the dangers we now face.
Antibiotic resistance is a naturally occurring phenomenon that can be slowed, but not stopped. Over time, bacteria adapt to the drugs that are designed to kill them, and change to ensure their survival. This makes previously standard treatments for bacterial infections less effective, and in some cases, ineffective.
Certain actions may accelerate the emergence and spread of antibiotic-resistant bacteria, such as:
Using or misusing antibiotics
Having poor infection prevention and control practices
Living or working in unsanitary conditions
Mishandling food
To protect yourself from harmful bacteria, wash your hands often with soap and water, or use an alcohol-based hand sanitizer. Healthy lifestyle habits, such as eating a proper diet, proper food handling, getting enough exercise and establishing good sleeping patterns, can also minimize the risk of illness.
You can also help tackle antibiotic resistance by:
Using antibiotics as directed and only when needed
Completing the full treatment course, even if you feel better
Never sharing antibiotics with others
Never using leftover prescriptions

Ever heard of PREDICTIVE MICROBIOLOGY?
Predictive microbiology is a branch of microbiology that entails that responses of microorganisms are reproducible as long as environmental factors affecting their growth are known..
Hence mathematical models are used to predict the growth response of microbes to environmental factors.. it is used mainly in the food industries to predict the shelf life of food products given that the chemical and physiological component of the products are already known..
Population of microbes can be predicted along side time... Predictions can never be 100% due to inconsistent microbial responses and variations in growth media..
These mathematical models can be obtained from some software packages such as:
* ComBase
* Pathogen modeling program
*Sym'Previus..
Its quite interesting to know this....

NEGATIVE STAINING
main purpose of Negative staining is to study the morphological shape, size and arrangement of the bacteria cells that is difficult to stain. eg: Spirilla. It can also be used to stain cells that are too delicate to be heat-fixed.
It is also used to prepare biological samples for electron microscopy. It is used to view viruses, bacteria, bacterial flagella, biological membrane structures and proteins or protein aggregates, which all have a low electron-scattering power
Principle of Negative Staining
Negative staining requires an acidic dye such as India Ink or Nigrosin.
India Ink or Nigrosin is an acidic stain.
This means that the stain readily gives up a hydrogen ion (proton) and the chromophore of the dye becomes negatively charged. Since the surface of most bacterial cells is negatively charged, the cell surface repels the stain. The glass of the slide will stain, but the bacterial cells will not. The bacteria will show up as clear spots against a dark background.
Reagents of Negative Staining
India ink :Nigrosin (Nigrosin 100 gm/L, Formalin 5 ml/L in water)
Procedure of Negative Staining
1. Place a very small drop (more than a loop full, less than a free falling drop from the dropper) of nigrosin near one end of a well-cleaned and flamed slide.
2. Remove a small amount of the culture from the slant with an inoculating loop and disperse it in the drop of stain without spreading the drop.
3. Use another clean slide to spread the drop of stain containing the organism using the following technique.
4. Rest one end of the clean slide on the center of the slide with the stain. Allow the stain to spread across the edge of the spreader and swiftly move the spreader forward to create a thin smear.
5 Air-dry the thin smear.
6. Focus a thin area under oil immersion and View unstained cells which are gray in colour via the electron microscope....
