Impressive new preprint research demonstrates single molecule localization of 30 different protein targets in neurons, with a theoretically unlimited number of targets possible.

The authors accomplish this feat by introducing a variation of DNA-PAINT labeling that takes advantage of a “secondary label” that binds to the docking strand and imaging strand. Removal or blocking of the secondary label is performed once the target has been sufficiently imaged, allowing for the same imaging strand to be used for imaging the next target.
