Serious adverse-event reporting needs a working workflow.

For dietary supplements, qualifying serious adverse-event reports received through the address or telephone number on the label must be submitted to FDA within 15 business days. New medical information received within one year of the initial report is also subject to a 15-business-day deadline. FDA recommends applying the same timing to serious reports received through other channels.

Adverse-event reports and related records must be retained for six years. Establish intake, seriousness assessment, follow-up, escalation, submission, and recordkeeping procedures before an urgent case arrives.

Phase III studies build on preliminary evidence of effectiveness and generate additional safety and effectiveness information needed to evaluate overall benefit–risk and support labeling.

Study design is not one-size-fits-all. The population, control, endpoints, analysis, and quality controls should fit the product, indication, prior evidence, and primary objective.

Audience: Pharmaceutical, biotechnology, and drug-development sponsors

The U.S. LDT regulatory landscape changed significantly after FDA’s 2024 final rule. A federal district court vacated that rule on March 31, 2025, and FDA subsequently restored the prior regulatory text on September 19, 2025.

Laboratories and test suppliers should avoid relying on outdated implementation timelines. Current legal requirements, existing laboratory obligations, and FDA enforcement-policy considerations should be evaluated separately using current sources.

OTC monograph compliance is not a one-time launch check.

FDA may use administrative orders to add, remove or revise monograph conditions. Regulatory and R&D teams should:

✓ Monitor proposed, interim final and final orders.
✓ Assess potential effects on products, labeling and testing.
✓ Consider the OMOR pathway when an industry-requested change is appropriate.

FDA’s live portal posted a new final administrative order on September 10, 2026—another reminder that regulatory intelligence is a lifecycle activity.

Does your mixing sheet prove the finished product will still support its label claims?

Formula targets are only the starting point. Ingredient variation, processing losses, moisture shifts, and storage can move actual nutrient levels away from plan. For a product positioned as “complete and balanced,” the nutritional adequacy basis and life stage also matter.

Practical checks:
• Set justified nutrient targets and action limits.
• Distinguish dry-matter calculations from as-fed label guarantees.
• Account for process and stability losses using relevant data.
• Confirm release specifications and trend results through shelf life.

A disciplined formulation-to-release strategy can reduce rework, out-of-specification results, and avoidable label revisions. Contact ProConXol for formulation, specification, and ingredient-review support tailored to your animal-food product.

A compliant OTC active ingredient is only the beginning.

A complete monograph-fit review should assess:

✓ Active ingredient
✓ Strength
✓ Indication
✓ Route
✓ Dosage form
✓ Directions and warnings
✓ Required testing

Each condition should align with the current applicable OTC monograph and administrative orders. One mismatch can affect the regulatory pathway, labeling strategy or launch plan.

A CAPA is not just a form.

A practical CAPA framework should define the problem, contain and evaluate affected product, assess risk and scope, investigate root cause, implement appropriate actions, verify effectiveness, and document closure.

These seven stages are a quality-system framework—not a universal FDA checklist. Specific corrective-action duties depend on the event and applicable regulation. Part 111 addresses investigations, disposition decisions, corrective actions, and recurrence prevention for relevant deviations. Part 117 establishes specific corrective-action and correction requirements for preventive-control programs.

Need help closing recurring quality issues? Ask ProConXol to review your CAPA system.

Build a claims-to-evidence matrix before the protocol is locked. Map every proposed claim to its endpoint, measurement method, population, data source, acceptance criteria, and submission use. This exposes evidence gaps early and reduces avoidable study redesign.

ProConXol develops claims-to-evidence matrices and clinical-evidence strategies for drug, device, and IVD programs.

eSTAR Is a Template—Not Your Regulatory Strategy

eSTAR provides a structured way to prepare a medical-device submission, but the template cannot solve a weak strategy. Indications for use, risk analysis, testing, labeling, and substantial-equivalence rationale must be coherent before the submission package is assembled.

Preparing an FDA submission? Ask ProConXol to review the strategy and evidence package.

Is your GRAS position tied only to a substance name—or to the actual conditions of use?

For animal food, GRAS is use-specific. A conclusion should match the ingredient’s identity and manufacturing method, target species, intended purpose, maximum use level, total dietary exposure, and supporting safety evidence. A conclusion involving human food, another species, or a different use level does not automatically transfer to a proposed animal-food use.

Before making commercial commitments, ask whether the marketed material matches the assessed identity, whether exposure assumptions reflect the complete diet, and whether the evidence addresses the intended animals—and potential human exposure when food-producing animals are involved.

ProConXol helps ingredient developers and animal-nutrition companies pressure-test GRAS positions, identify evidence gaps, and organize the regulatory strategy around the proposed conditions of use. Contact ProConXol for an animal-food GRAS readiness assessment.
